Proofreading happens after layout, not before. That is the whole point — a proofreader reads the book as it will be printed, which means catching problems that only exist once type is set.
What a proofreader catches

Proofreading is a safety net over already-edited text. If a manuscript has never been copy edited, a proofread will not rescue it — we will tell you that rather than take the booking.
